No, adding a transmitter to the spare will not allow you to include it in the rotation without the light activating. From what I have read about the system in the service manual any new transmitter added to the system would require registering that transmitter so it can be read by the remote keyless entry module and BCM. You actually have to register all 4 when you replace one. For that reason having one in the spare would require you to take your truck to the dealer to activate that new transmitter every time you rotated your tires. That could prove to be expensive.
